Total intensity is the sum of the pixel intensities of every pixel in the region of interest (ROI). To determine this information, you need to create a new Custom Measurement in the Measurements table which is the result of area x mean, where

Area (uncalibrated document) = number of pixels Mean = sum of pixel intensities / number of pixels Total intensity = area*mean = (number of pixels)(sum of pixel intensities/ number of pixels)

therefore Total intensity = sum of pixel intensities

Keep in mind that the document that you are working with must be uncalibrated so that the unit for area is square pixels and not square microns. In uncalibrated documents, area is simply the number of pixels.
